چکیده

The mammalian GABAJbenzodiazepine (GABAJBZ) receptor is comprised of several subunit isoforms: cq6, f3.3, Y,.~ and 6. In the present studies, the expression of (Y,, p2, and y2 subunit mFiNAs was examined in cerebellar Purkinje cells and deep cerebellar neurons of staggerer mutant mice during postnatal development. In control animals, the three subunit mRNAs were present at high density in Purkinje cells which, in adult animals, form a monolayer at the interface of the granule cell and molecular layers. The number of Purkinje cells in the staggerer cerebellar cortex is reduced; the majority of those that remain are retained within the granule cell layer and are unable to receive normal afferent synapses from granule cells. The three subunit mRNAs were expressed at similar levels in both staggerer and control Purkinje cells until postnatal day 9. After this time, although the cy, subunit mRNA was maintained at control levels in staggerer Purkinje cells, the expression of p2 and y2 subunit mRNAs decreased, and was largely absent by postnatal day 20. The loss of p2 and y2 mRNA expression in staggerer was specific to Purkinje cells, since all three mRNAs were present throughout postnatal development in other brain regions, including the deep cerebellar nuclei. The present studies indicate that in cerebellar Purkinje cells, the GABAJBZ receptor cq, and p2, and yz, subunit mRNAs are regulated by distinct mechanisms which are differentially affected by the staggerer mutation. The Lurcher cerebellar mutant phenotype is not expressed on a staggerer mutant background.

The hierarchy of the various processes responsible for the development of the complex, elaborated Purkinje cell can be examined by taking advantage of a series of spontaneous mutations that affect cerebellar development in the mouse. This study uses double mutants containing genes for two separate hereditary cerebellar mutations that have been shown to act intrinsically in Purkinje cells in ord...

ATP-dependent glutamate uptake into synaptic vesicles from cerebellar mutant mice.

The ATP-dependent glutamate uptake system in synaptic vesicles prepared from mouse cerebellum was characterized, and the levels of glutamate uptake were investigated in the cerebellar mutant mice, staggerer and weaver, whose main defect is the loss of cerebellar granule cells, and the nervous mutant, whose main defect is the loss of Purkinje cells. The ATP-dependent glutamate uptake is stimulat...

Actions and release characteristics of secretin in the rat cerebellum

Secretin, a peptide hormone of the gastrointestinal system, has been implicated in the etiology of autism. Our laboratory previously demonstrated the expression of secretin and its receptors in specific central neurons, and found for the first time that secretin is neuroactive in the cerebellum. We showed that bath application of secretin facilitated the release of GABA from terminals of basket...
